چکیده

n engl j med 360;8 nejm.org february 19, 2009 813 medicines should be an overarching goal for pharmacotherapy over the next decade. Pharmacogenetics has the potential to increase benefit and reduce harm in people whose drug responses are not “average.” In some cases, randomized, controlled trials will be needed to determine whether pharmacogenetic testing is worthwhile; in others, less rigorous approaches will suffice. Given the expected volume of genetic information and the relative paucity of randomized, controlled trials involving marketed drugs, we need clear thinking about what is required for the adoption of pharmacogenetic testing.

Phosphoglycerate Dehydrogenase: Potential Therapeutic Target and Putative Metabolic Oncogene

Exemplified by cancer cells' preference for glycolysis, for example, the Warburg effect, altered metabolism in tumorigenesis has emerged as an important aspect of cancer in the past 10-20 years. Whether due to changes in regulatory tumor suppressors/oncogenes or by acting as metabolic oncogenes themselves, enzymes involved in the complex network of metabolic pathways are being studied to unders...

Decoding key nodes in the metabolism of cancer cells: sugar & spice and all things nice

In the past 5 years, a convergence of studies has resulted in a broad appreciation in the cancer research community that reprogramming of cellular metabolism may be more central to cancer than appreciated in the past 30 years. The re-emergence of cancer metabolism stems in part from discoveries that a number of common oncogenes and tumor suppressor genes more directly control cell metabolism th...

Oncogenes and Tumor Suppressors Regulate Glutamine Metabolism in Cancer Cells

Several hallmarks of cancer cells are their display of metabolic changes and enhanced proliferation. Highly proliferating cells utilize glutamine as a source of nitrogen, and therefore, one of the commonly seen metabolic changes is increased glutaminolysis, or glutamine catabolism. In addition, glutamine is an important anaplerotic source by which cells support the pools of TCA cycle intermedia...

Balancing glycolytic flux: the role of 6-phosphofructo-2-kinase/fructose 2,6-bisphosphatases in cancer metabolism

The increased glucose metabolism in cancer cells is required to fulfill their high energetic and biosynthetic demands. Changes in the metabolic activity of cancer cells are caused by the activation of oncogenes or loss of tumor suppressors. They can also be part of the metabolic adaptations to the conditions imposed by the tumor microenvironment, such as the hypoxia response. Among the metaboli...

Human MicroRNA Oncogenes and Tumor Suppressors Show Significantly Different Biological Patterns: From Functions to Targets

MicroRNAs (miRNAs) are small noncoding RNAs which play essential roles in many important biological processes. Therefore, their dysfunction is associated with a variety of human diseases, including cancer. Increasing evidence shows that miRNAs can act as oncogenes or tumor suppressors, and although there is great interest in research into these cancer-associated miRNAs, little is known about th...
